They can't. For Nato to come to the defense of a member state, all member countries of Nato must agree to it. In the unlikely scenario that the US invades Greenland, the US will vote against Nato fighting against the US.

But leave Trump and his loud mouth. He can't invade Greenland.

I honestly do not see that happening, it's an unlikely event. NATO forming an alliance with Russia, China, against the US is another unlikely event, aside the growing fears of Russia expansionist agenda among its neighbouring NATO Allies. But I feel like NATO are not being pragmatic for the fact that they had a secret meeting about it in the first place. I can't envisage a military confrontation between NATO and the US, however there could be a very strained relationship.
